A susceptible pneumococcal isolate is incubated with a cell-free preparation from a resistant strain. Rare recipient colonies acquire the donor resistance allele and retain it after repeated growth without antibiotic. Treating the preparation with DNase before incubation eliminates acquisition; RNase and protease do not. No donor cells or infectious phages are detected.
